What the research says
Shiitake's signature compound is lentinan, a beta-glucan that's been studied since the 1980s and is used clinically in parts of Asia as an immune adjuvant. The mechanism is well-characterized: beta-glucans like lentinan bind to receptors on immune cells, including dectin-1 and toll-like receptors, and modulate the immune response rather than crudely “boosting” it.
The standout is a 2015 randomized human trial. Fifty-two healthy adults ate whole shiitake daily for four weeks. The results were specific and measurable: a 60% increase in the proliferation of gamma-delta T cells and a two-fold increase in NK-T cells, both meaningful immune-cell populations, alongside a rise in salivary IgA, which points to better mucosal and digestive immunity, and a reduction in C-reactive protein, a marker of inflammation. That combination, stronger immune cell activity plus lower inflammation, from a food, in actual humans, over four weeks, is exactly the kind of evidence most “immunity” supplements can only gesture at. Shiitake has it.
